**How to acclimating your tissue culture after received plant:
When tissue culture plants are transferred from the lab to soil they are exposed to abiotic stresses, like altered temperature, light intensity, and humidity conditions, and biotic stresses, like soil microflora (microbes living in soil). So, they need step-wise acclimatization to successfully establish themselves in the natural environment.
1. Open the bottle/pack in a shaded, cool, and most importantly, clean environment.
2. Use a tweezer to gently take the tissue cultures out of the pack/glass bottle.
A useful technique is to open the bottle/flask and pour some lukewarm water into it. Swirl the flask so that the entire clump of plants, along with jelly, loosens up and starts to swirl with the swirling action.
Then carefully take this plant (use a tweezer) and gel clump into lukewarm water, and then carefully separate jelly from roots/plant’s base. This not only makes it easier to clean off all the gel, but the roots also tend to suffer less damage this way.
3. Soak them in anti-fungus for 10-15 mins (Betadine mix with water will also work)
Remove plantlets from only one flask at a time and plant them quickly.
4. Prepare another bowl of distilled water and a few drops of rooting hormone. Soak the tissue cultures in this mixture for 5 minutes.
5. Plant tissue cultures into a mixture of 5:5 [perlite and peat moss (or coco coir)]
6. Put them into the closed package (A clear plastic cup with a tight lid Or planting trays using plastic bags to cover baking.) – allowing light to pass through. Bright indirect light or under a grow light.
7. Acclimate for 2-4 weeks with LED lighting for 12-18 hr/day.
Temperatures within a range of 18-23°C.
Maintaining humidity at around 70% – 90%.
